Schizophrenia (SCZ) is a devastating disorder with no known cause, nor any specific treatment available. However, with the recent identification of specific gene variants, it is possible to identify the gene- environment interactions which could help in identifying the pathophysiology of the disease. We used a NIMH-sponsored study to identify the genetic component of SCZ. Several independent studies involving genome-wide association (GWA) mapping have now identified markers that appear to be a part of genetic risk factors for SCZ. However, these markers have not replicated in any other GWA studies or studies conducted in different ethnic groups. Thus, the current consensus is that a substantial number of risk alleles exist for SCZ, but the functional significance of the associated markers is not known. A large number of SCZ patients are African-American. Thus, the current study also focuses on African-Americans. We will identify specific genetic risk factors for SCZ in African-American subjects, to understand the genetic basis of SCZ in this ethnic group, and to determine if the genetic risk factors can be useful to identify genes that might be involved in determining susceptibility to SCZ in African-Americans. This research may help to identify gene-environment interactions that may be important in the pathophysiology of the disease.
